Gastrointestinal motility regulatory targets represent a broad group of molecular entities, primarily G protein-coupled receptors and ion channels, that coordinate the mechanical movement of the digestive tract. These targets are located throughout the enteric nervous system and on gastrointestinal smooth muscle cells, where they coordinate peristalsis, segmentation, and secretion (Camilleri, 2019, PubMed). Key examples include the 5-HT4 receptor, which promotes acetylcholine release to stimulate contraction, and the dopamine D2 receptor, which provides inhibitory tone (Galligan & Vanner, 2005, Gastroenterology). Therapeutic agents targeting these molecules, such as prokinetics (e.g., prucalopride) or secretagogues (e.g., linaclotide), are used to manage disorders like gastroparesis and chronic constipation (Ford et al., 2014, American Journal of Gastroenterology). Conversely, antagonists like 5-HT3 blockers are used to slow motility in conditions like diarrhea-predominant irritable bowel syndrome (Sanger & Alpers, 2008, Nature Reviews Gastroenterology & Hepatology). Because these targets are often expressed in other tissues, drugs must be carefully designed to avoid systemic side effects such as cardiac arrhythmias or central nervous system disturbances (Blackshaw et al., 2007, Autonomic Neuroscience).
