The gastrointestinal tract barrier function refers to the physiological role of the gut wall in separating the internal milieu from the external environment, primarily via the mucus layer, intestinal epithelial cells joined by tight and adherens junctions, and the gut vascular barrier[1][2][4]. This barrier selectively permits the absorption of nutrients and water while blocking the passage of pathogens, toxins, and antigens. Its integrity is maintained by complex interactions between epithelial cells, immune cells in the lamina propria, commensal microbiota, and secreted proteins like mucins and antimicrobial peptides. Barrier dysfunction is implicated in a wide spectrum of gastrointestinal and systemic diseases, and various therapeutic approaches aim to restore its integrity by targeting these underlying molecular and cellular components[1][2][4].
