Gastrokine-1 (GKN1) is a highly conserved, protease-resistant, secreted protein encoded by the GKN1 gene, expressed predominantly in the gastric foveolar epithelial cells and restricted to the stomach mucosa. It maintains gastric mucosal integrity, regulates cell proliferation, differentiation, and repair, and suppresses inflammatory signaling pathways (e.g., NF-κB, COX-2). The BRICHOS domain mediates protein-protein interactions and is associated with suppression of cancer and prevention of amyloid aggregation. Gastrokine-1 has been implicated as a tumor suppressor, with loss of expression linked to the development and progression of gastric cancer.
