Overview

GATA binding protein 3 (GATA3) mRNA is the messenger RNA that encodes the GATA3 transcription factor, a master regulator of T-helper type 2 (Th2) cell differentiation and the production of pro-inflammatory cytokines such as IL-4, IL-5, and IL-13. In diseases like asthma, COPD, and ulcerative colitis, GATA3 is overexpressed, leading to excessive Th2-mediated inflammation and tissue damage. Therapeutic strategies targeting GATA3 mRNA involve the use of DNAzymes, such as SB-010 (hgd40), which are synthetic single-stranded DNA molecules designed to bind and catalytically cleave the GATA3 mRNA sequence. By reducing the levels of GATA3 mRNA, these drugs prevent the synthesis of the GATA3 protein, thereby modulating the underlying immune response and alleviating symptoms of allergic and inflammatory conditions. The name 'SB-101 mRNA' appears to be a confusion with the drug candidate SB-010 or the unrelated TLR9 agonist SB-101, as GATA3 mRNA is the primary mRNA target associated with the 'SB' series of inhaled oligonucleotides developed by Sterna Biologicals.
