GATA binding protein 6 (GATA6) is a member of the GATA family of zinc finger transcription factors, playing a pivotal role in the development and differentiation of endoderm- and mesoderm-derived tissues, including the heart, lungs, and gastrointestinal tract (UniProt Q92908). In oncology, GATA6 acts as a lineage-specific regulator; its amplification is linked to the progression of esophageal and gastric cancers, while its loss in pancreatic ductal adenocarcinoma (PDAC) marks a transition to a more aggressive, chemotherapy-resistant basal-like subtype (PubMed: 24317510, 35469023). Beyond cancer, GATA6 is a critical host factor for SARS-CoV-2, directly regulating the expression of the ACE2 receptor to facilitate viral entry (PubMed: 35469023). While direct small molecule inhibitors like Pyrrothiogatain are primarily used in research, GATA6 is a major focus for patient stratification and the development of RNA-based or indirect therapeutic strategies (PubMed: 31757955). Its complex role as both a potential oncogene and a tumor suppressor necessitates precise, context-specific targeting to avoid developmental or systemic toxicities.
