Overview

GATA zinc finger domain containing 2A (GATAD2A) is a protein-coding gene that encodes a subunit of the nucleosome remodeling and deacetylase (NuRD) complex, a crucial multi-protein complex involved in chromatin remodeling and transcriptional repression[1][5][7]. GATAD2A, also known as transcriptional repressor p66-alpha, functions as a protein-macromolecule adaptor within the NuRD complex, facilitating the negative regulation of DNA-templated transcription and gene silencing primarily through facilitating histone deacetylation[1][7]. It enhances MBD2-mediated repression and forms an efficient repressor complex in conjunction with GATAD2B. GATAD2A is localized to the nucleoplasm and is implicated in several cellular pathways, including DNA repair (homologous recombination) by recruiting the NuRD complex to sites of DNA damage[2]. Mutations and alterations in GATAD2A have been associated with cancer (notably sarcomas), dilated cardiomyopathy, and Sifrim-Hitz-Weiss syndrome, highlighting its relevance to disease but not as a direct drug target[1][2].
