GATA2 antisense RNA 1 (GATA2-AS1) is a long non-coding RNA transcribed antisense to the GATA2 gene, with its transcription start site located ~1.2kb from GATA2. It is highly expressed in endothelial cells and several highly vascularized tissues (lung, placenta, kidney), and is also upregulated in various cancer cell types, including colorectal and non-small cell lung cancer. GATA2-AS1 functions in regulatory feedback loops with the protein-coding GATA2 gene, stabilizing GATA2 mRNA and being transcriptionally activated by GATA2, thereby influencing critical pathways in cancer progression, especially epithelial-mesenchymal transition, migration, invasion, and maintenance of cancer stemness. In the endothelium, GATA2-AS1 coordinates metabolic adaptation and angiogenesis, especially in hypoxic conditions, by maintaining expression of glycolytic genes. While emerging evidence points to its role as a potential therapeutic target and biomarker in cancer and vascular biology, no approved drugs target this RNA directly.
