Overview

GATA3 antisense RNA 1 (GATA3-AS1) is a divergent long non-coding RNA located adjacent to the GATA3 gene, sharing its promoter. It is selectively expressed in TH2 cells and required for efficient transcription of GATA3 and key cytokine genes (IL5, IL13). GATA3-AS1 acts by recruiting MLL methyltransferase to its locus through an R-loop structure, facilitating chromatin modification and transcriptional activation under TH2 differentiation. Dysregulation of GATA3-AS1 has been implicated in various cancers, where it promotes cell proliferation, migration, invasion, immune evasion (via PD-L1 stabilization), and epithelial–mesenchymal transition. Elevated levels of GATA3-AS1 are associated with tumor progression and poor chemotherapy response. It is considered both a mechanistic driver and a potential biomarker in cancers and immune disorders

Mechanism of action

siRNA-mediated knockdown of GATA3-AS1 reduces GATA3 and downstream cytokine expression. Modulation of chromatin structure through interaction with MLL methyltransferase. Targeting GATA3-AS1 may impair tumor proliferation, migration, invasion, and immune escape.
