Overview

GATA6 antisense RNA 1 (GATA6-AS1) is a long non-coding RNA located adjacent to the GATA6 gene, which encodes a cardiac transcription factor[4]. GATA6-AS1 is transcribed from the antisense strand and is regulated by a bidirectional promoter shared with GATA6, leading to co-expression in human tissues, most notably in the heart, ovary, colon, and small intestine[4]. Functionally, GATA6-AS1 is involved in key regulatory roles, including control of endothelial gene expression via interaction with the epigenetic regulator LOXL2, thereby modulating histone methylation and angiogenesis[1]. In cardiac biology, GATA6-AS1 is necessary for proper cardiomyocyte differentiation from pluripotent stem cells, acting as a positive regulator of GATA6 expression and mesodermal lineage gene pathways[3][4]. In the gut, GATA6-AS1 maintains epithelial mitochondrial function and energy metabolism; its downregulation is associated with increased disease severity and worse clinical outcomes in ulcerative colitis, Crohn's disease, and coeliac disease[5]. GATA6-AS1 does not encode a protein and is not considered a classical therapeutic target such as a receptor, enzyme, or transporter; rather, it acts as a gene regulator in physiology and disease[1][5]. There are currently no drugs directly targeting GATA6-AS1, but its expression serves as a biomarker for disease monitoring and possibly for the identification of therapeutic windows in certain gut pathologies and cardiac development studies[5].
