Overview

GC-rich duplex DNA motifs, often called GC boxes, are essential regulatory sequences located within the promoter regions of a wide array of genes, including those responsible for cell growth, survival, and differentiation (PMID: 22431506). These motifs are the primary recognition sites for the Specificity Protein 1 (Sp1) transcription factor, which facilitates the recruitment of the basal transcription machinery (PMID: 11509596). In various cancers, the dysregulation of Sp1-mediated transcription leads to the overexpression of oncogenes such as VEGF, c-Myc, and Survivin, making these DNA sequences attractive therapeutic targets (PMID: 25633343). Small molecules like Mithramycin A and its analogues target these motifs by binding to the minor groove in a magnesium-dependent manner, effectively displacing Sp1 and inhibiting gene expression (PMID: 15155826). While effective in preclinical models, the clinical application of drugs targeting GC-rich DNA is often limited by significant toxicities, including hepatotoxicity and thrombocytopenia, due to the ubiquitous nature of these motifs across the genome (PMID: 21854183). Recent research focuses on developing analogues with improved therapeutic indices and greater specificity for particular promoter contexts.
