GDNF family receptor alpha-2 (GFRA2) is a glycosylphosphatidylinositol (GPI)-anchored cell surface receptor that preferentially binds the neurotrophic factor neurturin and also interacts with glial cell line-derived neurotrophic factor (GDNF). GFRA2 mediates the activation of the RET tyrosine kinase receptor and supports signaling pathways involved in the survival and differentiation of sympathetic and other neurons. The protein and its gene have been implicated in neurodevelopmental and neurodegenerative diseases as well as certain cancers, notably neuroblastoma. GFRA2 is distinct from its family members by its ligand specificity and physiological roles.
Ligand binding (especially neurturin): interaction with neurturin leads to complex formation with the RET tyrosine kinase, triggering downstream signal transduction and impacting neuronal survival
