GDP-fucose biosynthesis refers to the cellular processes that generate GDP-L-fucose, a critical sugar nucleotide donor used in the fucosylation of glycans. Fucosylated glycans are essential for various biological functions, including cell signaling and immune responses. There are two main pathways: the de novo pathway (from GDP-mannose via GMDS and TSTA3/FX) and the salvage pathway (from free L-fucose via FUK and FPGT). Regulation involves feedback inhibition and interplay between pathways. After synthesis in the cytosol, SLC35C1 transports UDP-Fuc/GDP-Fuc into the Golgi lumen. The enzymes involved are conserved across many species.
