Gem-interacting protein (GMIP) is an enzyme belonging to the ARHGAP (Rho GTPase activating protein) family. It interacts with the Ras-related GTPase Gem via its N-terminal, and with RhoA via its RhoGAP domain, stimulating the GTPase activity of RhoA in vitro and in vivo. GMIP regulates actin cytoskeletal reorganization and plays a role in neuronal structure development, neurite growth, and axonal guidance. Human GMIP is encoded by the GMIP gene (NCBI Gene ID: 51291), located on chromosome 19. Alternative names include ARHGAP46 and GEM-interacting protein. GMIP is regarded as an enzyme—specifically a Rho GTPase-activating protein—rather than a typical drug target such as a receptor, transporter, or kinase. There are currently no known drugs or standard mechanisms of drug action directly targeting GMIP in therapeutic settings, nor are there established clinically validated biomarkers or safety concerns linked to its modulation. Its main established biological role is in cytoskeletal organization and neuronal development, with limited evidence linking it to diseases such as major depressive disorder and possibly certain cancers, primarily via gene expression studies.
