Overview

Geminin coiled-coil domain-containing protein 1 is a member of the Geminin family of chromatin-binding proteins, encoded by GMNC on chromosome 3q28. It is a transcriptional regulator essential for the differentiation of multiciliated cells in diverse tissues, including the respiratory tract, brain, and reproductive system. GMNC/GEMC1 acts by activating transcriptional programs for multiciliogenesis, promoting expression of key regulators such as MCIDAS, FOXJ1, TP73, CCNO, and others that drive MCC fate and ciliogenesis. GMNC also interacts with proteins like E2F4/5-DP1, Multicilin (MCIDAS), and Geminin through its coiled-coil and TIRT domains, forming complexes that orchestrate gene activation during multiciliogenesis. Deficiency or mutation in GMNC/GEMC1 impairs MCC development and leads to ciliopathies such as Reduced Generation of Multiple Motile Cilia (RGMC), hydrocephalus, chronic airway mucus accumulation, and reproductive system infertility. There is no evidence that GMNC/GEMC1 is currently used as a therapeutic drug target or biomarker in clinical practice.
