General cellular uptake and the endosomal membrane compartment represent the collective biological processes and organelles involved in endocytosis and intracellular trafficking. This system is responsible for the internalization of extracellular ligands, nutrients, and membrane proteins, which are then sorted for recycling to the cell surface or degradation in lysosomes (Alberts et al., Molecular Biology of the Cell). While not a single molecular target, this pathway is a critical focal point for drug delivery, as many therapeutic modalities, such as antibody-drug conjugates and lipid nanoparticles, rely on endosomal uptake and subsequent escape to reach their functional sites (Sahay et al., Nature Biotechnology, 2013). Furthermore, numerous pathogens, including viruses like SARS-CoV-2 and influenza, exploit the endosomal compartment for host cell entry (Cano-Cano et al., International Journal of Molecular Sciences, 2023). Consequently, pharmacological modulation of endosomal pH or trafficking proteins is explored in treating infections and metabolic disorders, though the ubiquitous nature of these processes presents significant challenges for therapeutic selectivity.
