Eukaryotic translation initiation factor 2-alpha kinase 4 (EIF2AK4), also known as GCN2, is a serine/threonine-protein kinase that plays a critical role in the regulation of protein synthesis in response to various cellular stresses. It phosphorylates the alpha subunit of eukaryotic translation initiation factor-2 (eIF2α), leading to downregulation of global protein synthesis as an adaptive response to diverse cellular stresses. Mutations or dysregulation in EIF2AK4 have been associated with certain genetic disorders affecting blood vessels and may play roles in rare pulmonary diseases.
