Overview

General host cancer cell machinery is a broad, non-specific term referring to the collective biological processes and molecular components that sustain the growth, survival, and proliferation of malignant cells within a host (Hanahan & Weinberg, 2011). This encompasses fundamental cellular operations such as DNA replication, transcription, translation, and metabolic pathways that are often dysregulated or hijacked in cancer to support uncontrolled expansion (NCI, 2024). Because this term describes a complex system of pathways rather than a single molecular entity like a receptor or enzyme, it is not classified as a specific therapeutic target in modern drug discovery. Traditional cytotoxic chemotherapies, such as antimetabolites and microtubule inhibitors, interact with various components of this machinery to induce cell death or arrest the cell cycle (ACS, 2023). However, because these processes are also essential for the function of healthy cells, targeting the general machinery often results in significant systemic toxicity and a narrow therapeutic index.
