Target intelligence / Profile preview
General lipid metabolism pathways refer to the integrated network of biochemical reactions responsible for the synthesis, transport, and degradation of lipids, including fatty acids, triglycerides, and cholesterol (StatPearls, 2023). These pathways are fundamental for energy storage, the formation of cellular membranes, and the production of signaling molecules like steroid hormones (NIH, 2019). While the term itself describes a broad biological process rather than a single molecular target, it encompasses numerous specific enzymes, receptors, and transporters that serve as critical therapeutic targets. Dysregulation of lipid metabolism is a hallmark of metabolic syndrome and is strongly associated with the development of atherosclerosis, cardiovascular disease, and nonalcoholic fatty liver disease (PubMed, 2021). Pharmacological agents often target specific nodes within these pathways, such as HMG-CoA reductase for cholesterol lowering or PPAR receptors for triglyceride management (Mayo Clinic, 2023). Consequently, monitoring lipid profiles remains a cornerstone of clinical practice for assessing metabolic health and drug efficacy.
Drugs targeting components of these pathways act through various mechanisms, including the inhibition of rate-limiting enzymes like HMG-CoA reductase, activation of nuclear receptors such as PPAR-alpha, or the inhibition of intestinal cholesterol absorption and proprotein convertase subtilisin/kexin type 9 (PCSK9).
