Overview

General protein substrates is a broad classification used in biochemical research and pharmacological databases, such as ChEMBL, to describe a non-specific collection of proteins used to measure enzymatic activity (ChEMBL, 2024). This term does not refer to a single molecular target but rather serves as a functional category for proteins—such as casein, albumin, or hemoglobin—that undergo enzymatic modifications like proteolysis or phosphorylation in experimental assays (Sigma-Aldrich, 2023). In a physiological context, these substrates represent the diverse pool of proteins that are processed by enzymes to maintain cellular homeostasis and facilitate signal transduction (NCBI, 2022). Because it is a generic category and not a distinct biological entity, it is not considered a valid therapeutic target for drug discovery. Instead, drugs are developed to target the specific enzymes, such as proteases or kinases, that act upon these substrates to treat diseases like cancer or inflammatory disorders (PubMed, 2021). Consequently, while these substrates are essential for in vitro screening and characterizing enzyme kinetics, they do not possess direct clinical or diagnostic utility as drug targets themselves.
