Overview

A general tissue scaffold is a three-dimensional, biocompatible structure designed to serve as a template for tissue regeneration and repair. Unlike traditional molecular targets such as receptors or enzymes, a scaffold acts as a physical surrogate for the extracellular matrix (ECM), providing the necessary mechanical support and surface chemistry for cell attachment and growth (NIH, 2023). These structures can be engineered from natural materials like collagen or synthetic polymers like polycaprolactone, often featuring tailored porosity to allow for nutrient diffusion and vascularization (ScienceDirect, 2022). In clinical practice, scaffolds are frequently functionalized with bioactive agents, such as growth factors or antimicrobial peptides, to enhance biological outcomes in treating bone defects, severe burns, or organ damage (PubMed, 2021). The primary therapeutic challenge lies in balancing the scaffold's degradation rate with the pace of new tissue formation to ensure long-term structural and functional integration.
