General transcription factor IIE subunit 1 (GTF2E1), also known as TFIIE-alpha, is a protein component of the general transcription factor TFIIE complex, which is essential for transcription initiation by RNA polymerase II in eukaryotes[3][8][1][10]. GTF2E1 recruits the TFIIH complex to the transcriptional initiation complex and stimulates its C-terminal domain kinase and DNA-dependent ATPase activities, both of which are necessary for promoter clearance by RNA polymerase II[4][9][10]. This factor is ubiquitously expressed and functions as part of the basal transcription machinery in the nucleus. Disruption or altered regulation of GTF2E1 has been reported in certain rare genetic disorders and may be regulated in cancer, but it is not currently considered a direct therapeutic target[3][5][9].
