Overview

General transcription factor IIE subunit 2 (GTF2E2) encodes the beta subunit of the TFIIE complex, a critical element of the RNA polymerase II transcription initiation machinery[1][3][5]. TFIIE (a heterodimer of alpha and beta subunits) is required for recruiting TFIIH and enabling promoter clearance, thus allowing transcription to move from initiation to elongation. The beta subunit, encoded by GTF2E2, facilitates conformational changes and stimulates the XPB helicase in TFIIH to promote promoter opening and efficient transcriptional initiation[1][3]. Mutations in GTF2E2 underlie nonphotosensitive trichothiodystrophy (TTD6) and have been mapped to regions of genetic diseases. Aberrant overexpression has been linked to certain cancers and is implicated in cell proliferation via pathways such as AKT/ERK/mTOR[1]. GTF2E2 has also been functionally tied to the HIV life cycle and is involved in basal cellular transcriptional regulation across many tissues[3][5]. There are no currently approved targeted drugs or therapeutics for GTF2E2, though its disease associations and biomarker potential are subjects of ongoing research.
