General transcription factor IIH subunit 3 (GTF2H3)

Target
GTF2H3
Molecular classification
Transcription factor, Basal transcription factor, TFIIH subunit
01

Overview

General transcription factor IIH subunit 3 (GTF2H3) is a core component of the TFIIH multiprotein complex, which is essential for the initiation of transcription by RNA polymerase II as well as for nucleotide excision repair of DNA. The GTF2H3 (also called p34 or TFB4) subunit contributes to the structural and functional integrity of the TFIIH core complex, coordinating interactions necessary for unwinding DNA at transcription start sites and at DNA lesions targeted for excision repair. TFIIH as a whole is implicated in several human diseases when disrupted, primarily through other subunits, but GTF2H3 is required for the proper function of both gene expression and DNA repair pathways[1][2][3][4][6][10].

Mechanism of action

Not applicable; no drugs are known to target GTF2H3 directly.

03

Biological functions

Gene transcription initiation (by RNA polymerase II)Nucleotide excision DNA repair
04

Disease associations

Cancer susceptibility (via genetic polymorphisms in TFIIH complex subunits)[1]Premature aging syndromes (by analogy with other TFIIH mutations)DNA repair deficiency diseases (e.g. trichothiodystrophy, xeroderma pigmentosum, mostly caused by other TFIIH subunits but the biological pathway is shared)[1]
05

Safety considerations

No direct drug safety concerns; mutation in TFIIH subunits can cause disease states (see above).
06

Interacting drugs

None known that bind GTF2H3 subunit specifically; TFIIH complex as a whole can be affected by inhibitors (e.g., triptolide targets XPB subunit)[1]
07

Biomarkers

None established for clinical use.
