General transcription factor IIH subunit 4 (GTF2H4) is a protein component of the TFIIH complex, which plays critical roles in both basal transcription by RNA polymerase II and nucleotide excision repair (NER) of DNA[1][4][6]. Within transcription, it's required for promoter opening and escape after the formation of the pre-initiation complex. In DNA repair, GTF2H4 (as part of TFIIH) helps unwind DNA around lesions, enabling excision of damaged nucleotides. The GTF2H4 subunit is essential for proper TFIIH complex structure and function, stimulates ATPase activity of XPB (another TFIIH subunit), and mediates interactions with other basal transcription factors like GTF2F1 and XPB[1][2][6]. Mutations are linked to rare repair-deficiency disorders such as trichothiodystrophy and Cockayne syndrome[1]. As of now, it is not a druggable target, but it is a key molecule in the fundamental cell processes of transcription and repair.
