Overview

General transcription factor IIIC subunit 3 (GTF3C3) is a core component of the transcription factor IIIC (TFIIIC) complex, a conserved eukaryotic six-subunit protein complex that is essential for the recruitment of the TFIIIB complex and for the initiation of RNA polymerase III (Pol III)-mediated transcription[1][2][6]. The TFIIIC complex functions by binding to specific promoter elements within small nuclear and cytoplasmic RNA genes, such as tRNA and 5S rRNA genes, facilitating pre-initiation complex assembly, and determining the transcription start site[1][6][7]. GTF3C3, as part of this complex, plays a vital role in these processes and also participates in chromatin organization and regulation of gene expression, including that of certain RNA polymerase II genes[1]. Biallelic variants in GTF3C3 have been associated with a neurodevelopmental disorder characterized by microcephaly, global developmental delay, intellectual disability, cerebellar atrophy, and seizures in humans and corresponding phenotypes in animal models, underscoring its critical role in neural development and gene regulation[2][4].
