Overview

General transcription factor IIIC subunit 4 (GTF3C4) is a protein component of the transcription factor IIIC (TFIIIC) complex, which is essential for the transcription initiation of small RNA genes by RNA polymerase III, such as 5S rRNA and tRNA genes[2][6][7][10]. GTF3C4 functions as a DNA-binding subunit and possesses histone acetyltransferase activity, specifically acetylating histone H3 at lysine 14 (H3K14), suggesting a role in chromatin remodeling and gene activation[2][9][10]. It interacts with other TFIIIC subunits (such as GTF3C1, GTF3C2, GTF3C5) and with components of RNA polymerase III machinery, facilitating the assembly of the transcription machinery at Pol III promoters[2][5][10]. Mutations or altered expression of GTF3C4 have been linked to several diseases, including certain cancers and psoriasis, but it is not currently considered a typical therapeutic target or druggable protein[2][7][9].
