General transcription factor IIIC subunit 5 (GTF3C5) is a component of the multi-subunit transcription factor IIIC complex, which is essential for RNA polymerase III-mediated transcription. This protein directly binds DNA sequence motifs in promoters of tRNA and other small RNAs, guiding RNA polymerase III to the initiation site. GTF3C5 is considered a tightly associated, integral element of the TFIIIC2 subcomplex and plays a crucial role in the transcriptional regulation of genes involved in cellular differentiation and ribosomal biosynthesis. It localizes to the nucleus and interacts with other TFIIIC complex proteins, including GTF3C2 and GTF3C4. Disease roles are limited and largely associative, with minimal evidence for direct involvement in major human diseases. No known drugs interact directly with this protein, and it does not serve as a biomarker or safety concern in clinical settings.
