Overview

Genomic DNA guanine bases are one of the four primary nucleobases that constitute the genetic blueprint of living organisms. Guanine is a purine derivative that pairs with cytosine through three hydrogen bonds, providing structural stability to the DNA double helix (NIH, 2023). In the context of pharmacology, guanine is a critical therapeutic target because its N7 and O6 positions are highly nucleophilic, making them susceptible to attack by electrophilic cytotoxic drugs (PubChem, 2024). Platinum-based agents like cisplatin and alkylating agents like temozolomide form covalent bonds with these guanine sites, creating DNA adducts that stall replication forks and inhibit transcription (StatPearls, 2023). This mechanism is widely exploited in oncology to induce apoptosis in malignant cells. Additionally, guanine-rich regions can form G-quadruplex structures, which are increasingly studied as targets for stabilizing telomeres or regulating oncogene expression (PubMed, 2021). Despite their efficacy, drugs targeting guanine bases often lack specificity, resulting in systemic toxicities and potential mutagenic effects in non-cancerous tissues.
