Geranylgeranyl pyrophosphate synthase (GGPPS) is an enzyme that catalyzes the synthesis of geranylgeranyl pyrophosphate (GGPP) from farnesyl diphosphate (FPP) and isopentenyl diphosphate (IPP). GGPP is a 20-carbon isoprenoid molecule essential for the post-translational modification of proteins via geranylgeranylation and as a precursor for vitamin K2, ubiquinone, carotenoids, gibberellins, tocopherols, and chlorophylls. Dysregulation or mutations in GGPS1 are linked to several human diseases, including cancer and pulmonary fibrosis. GGPPS inhibitors are under investigation as potential therapeutics.
