Overview

Geranylgeranyltransferase type I (GGTase-I) is an essential eukaryotic enzyme that catalyzes the posttranslational modification of proteins by transferring a geranylgeranyl group to the cysteine residue within a C-terminal CaaX motif of target proteins. This facilitates membrane association and proper function of numerous signaling proteins, especially those involved in cell regulation and oncogenesis. GGTase-I is a heterodimer composed of two subunits: alpha (FNTA gene) and beta (PGGT1B gene). It modifies more than 100 different proteins, including many small GTP-binding regulatory proteins such as members of the Rho family and some Ras-related proteins. Due to its role in cancer progression, inhibition of GGTase-I has been explored as an anti-cancer strategy.
