Overview

GCNA is an evolutionarily conserved, germline-enriched protein characterized by a highly acidic, intrinsically disordered region and a protease-like SprT/minigluzincin domain. GCNA is essential for maintaining genome stability in germ cells, facilitating DNA repair, chromosome segregation, and cell cycle control, and preventing harmful DNA-protein crosslinks that would threaten fertility and propagational integrity. Homology with Wss1 and Spartan proteins places GCNA in a small family of IDR-containing metalloproteases specialized for cells with heritable genomes. Mutations in GCNA impair fertility and result in germ cell developmental defects and genomic instability, making it a key research marker for reproductive biology and germ cell cancer studies.
