Overview

Germ cell-specific gene 1-like protein (GSG1L) is a claudin-type transmembrane protein that serves as an auxiliary subunit of the AMPA (α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid) receptor complex[2][4]. It is expressed late during brain development in a region- and cell-type-specific pattern, constituting a small subset (about 5%) of AMPAR complexes in adulthood[2]. GSG1L acts postsynaptically to modulate AMPA receptor gating kinetics, specifically suppressing AMPA receptor-mediated synaptic transmission and altering short-term synaptic plasticity by promoting rapid AMPAR desensitization and deactivation[1][4]. Loss of GSG1L can lead to increased neuronal excitability and enhanced seizure susceptibility, as well as cognitive deficits in animal models[1][4]. While GSG1L is not a direct drug target as of current knowledge, its unique molecular and physiological properties distinguish it from other AMPAR auxiliary proteins (like TARPs and CNIHs), and it plays an essential role in fine-tuning excitatory neurotransmission and network plasticity in the brain[2][4].
