Overview

Germinal center-associated signaling and motility-like protein (GCSAML), also known as C1orf150, is a poorly characterized adaptor protein named for its sequence similarity to GCSAM, a protein involved in B cell receptor signaling[5][1][4]. GCSAML is predicted to contain two immunoglobulin-like domains and is primarily expressed in germinal centers (sites of B cell proliferation and differentiation), hematopoietic stem and progenitor cells, some myeloid/lymphoid lineages, and is highly enriched in mast cells in primates[1][3][4]. It likely functions in cell signaling within immune cells, especially in mast cell activation pathways, interacting with the FcεRI and KIT receptors, and being subject to tyrosine phosphorylation and myristoylation[2][4]. GCSAML is not a classical drug target (such as a receptor or enzyme) and no approved drugs are known to interact with it[1][3][4][5]. Genome-wide association studies (GWAS) and functional genomics have implicated splice variants of GCSAML (notably rs56043070[A] causing GCSAML-Δexon2) as genetic risk factors for urticaria, potentially affecting mast cell degranulation[5][2][4]. There is no conclusive evidence that GCSAML is currently pursued as a direct therapeutic target, but it acts as a putative immune modulator with potential biomarker applications.
