Overview

Germinal center kinases (GCKs) are a large subfamily of serine/threonine kinases in the STE20 family, highly conserved across eukaryotes and widely expressed in mammalian tissues. They are critical regulators of multiple cellular processes, including signal transduction, apoptosis, immune response, cell cycle progression, cell proliferation, migration, and cytoskeletal dynamics. GCKs participate in key signaling pathways involving MAP kinases (ERK, JNK, p38), influence both innate and adaptive immunity, and play context-dependent roles in health and disease, particularly in cancer, autoimmune disease, and inflammation. Members of the family include multiple kinases with diverse but overlapping functions, such as GLK (MAP4K3), TNIK, STK24, STK25, and others.
