Gigaxonin (GAN), also known as Kelch-like protein 16 (KLHL16), is an adaptor protein for Cul3-E3 ubiquitin ligases. It plays a crucial role in regulating the turnover of intermediate filament (IF) proteins, including neurofilaments, vimentin, keratins, and GFAP. Mutations in GAN cause giant axonal neuropathy (GAN), characterized by abnormal accumulation of neurofilaments. Recent research suggests a potential role as a tumor suppressor in head and neck cancers.
