Overview

Glial cell line-derived neurotrophic factor family receptor alpha (GFRα) refers to a family of glycosylphosphatidylinositol (GPI)-anchored cell surface receptors that serve as co-receptors for the GDNF family ligands (GFLs), including glial cell line-derived neurotrophic factor (GDNF), neurturin, artemin, and persephin. Upon ligand binding, the complex recruits and activates RET tyrosine kinase receptor, triggering downstream signaling pathways that promote neuronal survival, differentiation, growth, and maintenance. GFRα receptors are involved in various biological processes including neuronal development, kidney development, and spermatogenesis. Abnormal expression or activation can contribute to tumorigenesis and developmental disorders.

Mechanism of action

Ligand binding to GFRα recruits and activates the RET tyrosine kinase receptor, triggering downstream signaling pathways such as MAPK/ERK and PI3K/AKT to promote neuronal survival, differentiation, growth, and maintenance. Can also interact with NCAM to activate alternative signaling cascades independent of RET activation.
