Overview

The GFRα1–RET receptor complex is a multi-component signaling unit essential for the development and maintenance of various neuronal populations and the kidney (Airaksinen & Saarma, 2002, Nat Rev Neurosci). It is primarily composed of the glial cell line-derived neurotrophic factor (GDNF) receptor alpha 1 (GFRα1), a GPI-anchored protein (UniProt P56159), and the Rearranged during Transfection (RET) receptor tyrosine kinase (UniProt P07949). Upon binding of the ligand GDNF to GFRα1, the complex recruits RET, leading to its dimerization and autophosphorylation of intracellular tyrosine residues. This activation triggers downstream signaling pathways such as MAPK/ERK and PI3K/Akt, which regulate cell growth and survival (Mulligan, 2014, Nat Rev Cancer). In oncology, aberrant activation of the RET component through genetic fusions or point mutations is a key driver in medullary thyroid carcinoma and certain non-small cell lung cancers (Subbiah et al., 2020, JCO). Conversely, the complex is a target for regenerative medicine, where GDNF-mediated activation is explored for the treatment of Parkinson's disease to promote dopaminergic neuron survival (Whone et al., 2019, Brain). Therapeutic strategies include highly selective RET inhibitors like selpercatinib and pralsetinib for cancer treatment (FDA, 2020), and GDNF mimetics or gene therapies for neuroprotection. Understanding the structural assembly of this complex has been crucial for developing drugs that can specifically modulate its signaling depending on the clinical context.
