Overview

Glial cell line-derived neurotrophic factor receptor alpha1 (GFRα1) is a glycosylphosphatidylinositol (GPI)-anchored cell surface receptor that acts as the primary co-receptor for glial cell line-derived neurotrophic factor (GDNF), a member of the transforming growth factor-β superfamily[1][2][3][5]. GFRα1 binds extracellular GDNF and facilitates activation of the RET receptor tyrosine kinase, triggering intracellular signaling pathways that promote neuronal survival, differentiation, axon growth, and synaptogenesis[1][2][5]. This receptor is essential for the development and maintenance of several neuronal populations, most notably midbrain dopaminergic neurons, and is also implicated in non-neural systems such as kidney morphogenesis and spermatogenesis[4]. GFRα1 is anchored specifically to the outer leaflet of the plasma membrane via a GPI anchor and resides predominantly in lipid rafts, where it coordinates with RET to regulate signaling intensity and specificity[2]. In certain contexts, GFRα1 can also mediate GDNF signaling independently of RET via NCAM or engage with heparan sulfate proteoglycans and other co-receptors[4][6]. Therapeutically, modulation of the GDNF-GFRα1 system is under investigation for neurodegenerative diseases such as Parkinson's disease and epilepsy, but delivery and safety challenges remain significant barriers to clinical translation[2].
