Overview

GLIPR1-like protein 1 (GLIPR1L1) is a member of the CAP (Cysteine-rich secretory proteins, Antigen 5, and Pathogenesis-related 1) superfamily and is primarily characterized in reproductive biology. It is a membrane-associated protein enriched in the testis and spermatozoa, where it plays a critical role in mammalian fertilization, particularly at the stage of sperm-oocyte membrane fusion. GLIPR1L1 interacts with IZUMO1—another essential sperm protein—to form high-molecular weight protein complexes involved in the adhesion and membrane fusion events necessary for fertilization. The protein is involved in acrosome reaction processes and localizes to specific regions of the sperm head. GLIPR1L1’s membrane anchoring may vary between species (e.g., GPI-anchored in some, direct membrane association in others). There are no known drugs or inhibitors targeting this protein, and it is not considered a therapeutic target. There is no evidence linking GLIPR1L1 directly to human disease outside reproduction.
