Overview

Global paracrine and regenerative pathways refer to the collective signaling mechanisms through which cells, particularly mesenchymal stem cells (MSCs), secrete bioactive factors to promote tissue repair and modulate immune responses (Gnecchi et al., 2008, PubMed: 18923564). Rather than direct differentiation into damaged tissue, these pathways involve the release of a 'secretome' consisting of growth factors like VEGF and HGF, cytokines, and extracellular vesicles that activate endogenous repair processes (Caplan & Correa, 2011, PubMed: 21337614). These pathways are critical in treating conditions such as myocardial infarction, where they reduce apoptosis and stimulate angiogenesis (Baraniak & McDevitt, 2010, PubMed: 20888334). While not a single molecular target, they represent a therapeutic strategy in regenerative medicine aimed at harnessing the 'injury drugstore' effect of stem cells. Challenges include the complexity of the secretome and potential safety concerns like unwanted pro-inflammatory responses or tumorigenicity (Teixeira et al., 2013, PubMed: 23621371).
