The globin locus regulatory element, or locus control region (LCR), is a long-range cis-regulatory DNA element that controls the expression of globin genes, particularly within the β-globin gene cluster. The LCR is essential for achieving high-level, tissue-specific expression of these genes in erythroid cells. It consists of several DNase I hypersensitive sites and enhances expression of linked globin genes at physiological levels. Its activity is highly tissue-specific, selectively activating β-globin gene transcription in erythroid cells. Disruption leads to diseases such as β-thalassemia.
