The glomerular filtration barrier (GFB) is a highly specialized, multilayered structure in the kidney that serves as the primary interface for blood filtration. It allows water and small to midsized solutes to pass from the bloodstream into Bowman’s capsule while restricting larger molecules, particularly proteins, thus playing a critical role in urine formation and maintaining plasma protein homeostasis. It consists of fenestrated endothelial cells, the glomerular basement membrane (GBM), and podocytes with slit diaphragms. Disruption of the GFB leads to proteinuria and kidney diseases.
