Overview

Glucocorticoid-induced transcript 1 protein (GLCCI1) is an intracellular protein encoded by the GLCCI1 gene, highly expressed in glomerular podocytes and induced by glucocorticoids. GLCCI1 plays a role in the development and maintenance of podocyte foot processes, crucial for the glomerular filtration barrier. It also acts as an anti-apoptotic mediator in thymic T cells during glucocorticoid exposure. Genetic variants in GLCCI1 have been associated with variability in the response to inhaled glucocorticoid therapy in asthma patients. GLCCI1 is not classified as a classical therapeutic drug target but may serve as a biomarker for kidney health and glucocorticoid therapy responsiveness. Its broader biological function and mechanistic involvement in disease remain to be fully characterized

Mechanism of action

Modulation of apoptosis in thymic T cells via anti-apoptotic activity induced by glucocorticoid-receptor signaling. Possible involvement in podocyte foot process formation and maintenance through transcriptional regulation and PI3K pathway association.

03

Biological functions

Regulation of podocyte development and maintenance, important for glomerular filtration barrier integrityAnti-apoptotic mediator in thymic T cells under glucocorticoid regulationEarly marker of glucocorticoid-induced apoptosis in immune cells
04

Disease associations

Glomerular diseases (including nephrotic syndrome, proteinuria, diabetic nephropathy)Response to glucocorticoid therapy, particularly in asthma (genetic variants associated with differential drug response)Potential involvement in certain cancers via fusion proteins (e.g., GLCCI1-BRAF and ST3GAL3-GLCCI1 fusions)
05

Safety considerations

No direct safety concerns specific to GLCCI1 modulation currently knownPotential therapeutic challenges include poorly understood function and the risk of off-target effects given the protein’s expression in multiple tissues
06

Interacting drugs

Indirect interaction with glucocorticoids (e.g., dexamethasone), as GLCCI1 is induced by glucocorticoid treatment

07

Biomarkers

Genetic polymorphisms in GLCCI1 (notably rs37973) correlate with reduced improvement in lung function after inhaled glucocorticoid therapy in asthma patients (serving as a pharmacogenomics biomarker for therapy response)GLCCI1 levels as marker of podocyte health and integrity in kidney disease models
