Gluconokinase (IDNK) is an enzyme that catalyzes the phosphorylation of D-gluconate to 6-phospho-D-gluconate using ATP as a phosphate donor, generating ADP as a byproduct. It belongs to the kinase family, transferring a phosphate group specifically to carbohydrate substrates and participating in the pentose phosphate pathway, an alternative glucose metabolism pathway critical for nucleotide and amino acid biosynthesis. The human gene is found on chromosome 9, with the best-established activity predicted to be gluconokinase and involvement in carbohydrate metabolic processes. While orthologs are well-characterized in bacteria (such as E. coli idnK), the role of human gluconokinase is less well defined, with no established connection to common disease targets, known drugs, or biomarker applications. The enzyme acts on the metabolic level but is not currently considered a major therapeutic drug target.
