Overview

Glucosamine is an endogenous amino monosaccharide (2-amino-2-deoxy-D-glucose) that serves as a fundamental building block for the biosynthesis of glycosaminoglycans and proteoglycans—the key components of cartilage extracellular matrix. In cartilage, glucosamine is crucial for the synthesis of hyaluronic acid, chondroitin sulfate, and keratan sulfate, thus contributing to the strength, elasticity, and lubrication of articular cartilage. Glucosamine supplementation is widely studied and used in therapies aimed at slowing cartilage breakdown and reducing pain in osteoarthritis, although clinical evidence on its efficacy is mixed. Glucosamine acts in part by supporting anabolic cartilage metabolism and exerting anti-inflammatory effects within the joint.
