Glucose-6-phosphatase catalytic subunit 1 (G6PC1) is an integral membrane protein of the endoplasmic reticulum that catalyzes the hydrolysis of D-glucose 6-phosphate to D-glucose and inorganic phosphate. This reaction is the final step in both gluconeogenesis and glycogenolysis, making G6PC1 a key regulator of glucose homeostasis. Variants in G6PC1 cause Glycogen Storage Disease Type Ia (GSDIa; von Gierke disease). Overexpression or dysregulation may contribute to altered hepatic glucose output seen in diabetes mellitus.
