Overview

Glucose-dependent insulinotropic polypeptide (GIP), also known as gastric inhibitory polypeptide, is a 42-amino acid peptide hormone that acts as an incretin. It stimulates insulin secretion from pancreatic beta cells in a glucose-dependent manner. It also modulates glucagon secretion and promotes adipogenesis. GIP exerts pleiotropic effects beyond glycemic control including promoting growth and survival of pancreatic beta-cells, influencing brain function, bone metabolism, cardiovascular system regulation, and gastrointestinal tract physiology. Targeting the GIP-GIPR axis is being investigated for treating type 2 diabetes mellitus (T2DM) and obesity.
