Overview

The Glucose oxidase – copper metal-organic framework (GOx-Cu MOF) interface is a synthetic biocatalytic architecture used in nanomedicine and biosensing, rather than an endogenous biological target. This interface involves the immobilization of the enzyme glucose oxidase (GOx) onto or within a copper-based metal-organic framework, which serves as a porous, catalytic carrier. In therapeutic applications, particularly oncology, the GOx component catalyzes the oxidation of glucose to gluconic acid and hydrogen peroxide (H2O2), effectively starving tumor cells of their primary energy source (He et al., 2020). The copper nodes within the MOF then act as nanozymes, facilitating a Fenton-like reaction that converts the produced H2O2 into highly reactive hydroxyl radicals (.OH), which induce localized oxidative damage and cell death (Liu et al., 2019). This dual-action approach is known as synergistic starvation and chemodynamic therapy (SCT). Beyond therapy, this interface is a critical component in electrochemical biosensors, where the synergistic catalytic activity allows for the sensitive and selective detection of glucose levels in clinical samples (Wang et al., 2021).

Mechanism of action

The system operates through a tandem catalytic process where glucose oxidase depletes local glucose (starvation therapy) and produces hydrogen peroxide, which is subsequently converted by the copper MOF into toxic hydroxyl radicals via a Fenton-like reaction (chemodynamic therapy).
