Overview

Glucose-regulated protein 78 kDa (GRP78, also known as Binding immunoglobulin protein [BiP] or Heat shock 70 kDa protein 5 [HSPA5]) is a major endoplasmic reticulum chaperone belonging to the HSP70 family. It plays a central role in facilitating proper folding and assembly of newly synthesized proteins within the ER lumen, managing misfolded proteins through the unfolded-protein response pathway, regulating apoptosis under cellular stress conditions, and modulating immune responses. Its upregulation is associated with various pathological states including cancer progression, autoimmune disorders such as rheumatoid arthritis, cardiovascular diseases like dilated cardiomyopathy and atherosclerosis, neurodegenerative diseases including Alzheimer’s disease and stroke-related injury, metabolic syndromes such as obesity/diabetes/pancreatitis, and infections where it can be hijacked by viruses for replication. Due to these diverse roles—especially its involvement in tumor survival mechanisms—GRP78 has emerged both as a therapeutic target across multiple indications and as an important biomarker for diagnosis/prognosis.[1][2][3][4]
