Target intelligence / Profile preview

Glucose transporter 5 (GLUT5)

Target
GLUT5
Molecular classification
Transporter, Major facilitator superfamily (MFS) transporter, Sugar transporter
01

Overview

Glucose transporter 5 (GLUT5), also known as SLC2A5, is a membrane protein belonging to the major facilitator superfamily (MFS) of transporters. Unlike other GLUT family members, GLUT5 exhibits a high specificity for fructose rather than glucose and is predominantly expressed in the small intestine, kidney, and sperm. It plays a crucial role in the absorption of dietary fructose from the intestinal lumen into enterocytes. Structurally, GLUT5 comprises twelve transmembrane domains forming two bundles and contains regions responsible for high-affinity fructose binding and transport. The transporter has been implicated in diseases linked to altered fructose metabolism and is increasingly investigated as a target in cancer and metabolic disorders[1][5][2][3].

Mechanism of action

Inhibition of fructose transport by blocking or modulating the GLUT5 transporter

03

Biological functions

Fructose transportDietary fructose absorptionCellular fructose uptake
04

Disease associations

CancerMetabolic diseaseOther (implicated in fructose-linked pathologies such as diabetes, obesity, and nonalcoholic fatty liver disease)
05

Safety considerations

The selective inhibition of GLUT5 could potentially affect normal fructose absorption, leading to gastrointestinal side effects or metabolic disturbances. Long-term effects of targeting this transporter in humans remain unknown[1].
06

Interacting drugs

There are currently no approved drugs specifically targeting GLUT5; research probes and inhibitor molecules for GLUT5 are under investigation[1].
07

Biomarkers

Overexpression of GLUT5 is considered a biomarker for certain cancers (e.g., breast cancer) and diseases related to altered fructose metabolism[1].
